Reasons for choosing the GFCC′ face:
- It's the PD-1 contact face, so blocking it is functionally meaningful
- Sufficiently solvent-exposed
- Not masked by glycans (N35, etc.)
- Human/mouse contact residues are mostly conserved, so cross-reactivity is plausible
Avoided the flat, hydrophilic back face — binders don't attach well there.
